The antibody produces a strong biotin-streptavidin/HRP staining at a 1/1000 - 1/2000 dilution in rat anterior pituitary. Staining is completely eliminated by pretreatment of the diluted antibody with 10-6 M of Beta-Endorphin. Pre-adsorption of the diluted antibody with 10-6M of the following substances had no effect on Beta-Endorphin labeling: methionine enkephalin, leucine enkaphalin, dynorphin A, dynorphin B, gamma-endorphin, alpha-endorphin, ACTH and alpha-melanocyte stimulating hormone.
Photo Description: IHC images of beta-endorphin staining in the rat anterior pituitary with FITC-conjugated secondary antibody (above) and with standard biotin-streptavidin/HRP and DAB chromogen (below). The tissue was fixed with 4% formaldehyde in phosphate buffer, before being removed and prepared for frozen sectioning. Sections were incubated at RT in 10% goat serum in PBS, before standard IHC procedure. Primary antibody was incubated at 1:2000 for 48 hours, goat anti-rabbit secondary was subsequently added for 1 hour after washing with PBS.
